Pronuclear Microinjection Services

  1. Purified, linearized DNA at a concentration of 4 ng/µl will be microinjected into one-cell fertilized eggs.
  2. Fertilized eggs will be obtained by mating (C57BL/6 X C3H)F1 or (C57BL/6 X SJL)F1 females to (C57BL/6 X C3H) F1 or C57BL/6 X SJL)F1 males, respectively. Eggs obtained from other strains of mice are available upon consultation with the facility.
  3. The fertilized eggs will be injected and then incubated overnight to access toxicity of the construct. Eggs dividing to the two-cell stage will be implanted in pseudopregnant foster mothers.
  4. Prioritization of DNA injection is by date of receipt of the DNA sample, however, clients submitting multiple samples will have one construct injected per week, to prevent a backup of samples from other clients.
  5. Clients will be notified by e-mail when their constructs have been microinjected.
  6. For efficient scheduling, it is helpful to provide the VARI transgenic core facility with advance notice of sample submissions.
