How to Submit Samples

Please see the items below to prepare and send your samples. If you will need a large scale genomic sequencing (cosmid/PAC/BAC clones), please contact us before you start preparing your DNA so we can schedule your project accordingly.

  • Pipet DNA templates and primers into either 96 well PCR plates or (with a smaller number of samples) into Strip-Ease PCR tubes (Robins Scientific #1044-50-0 through 1044-50-6, caps 1044-72-0 through 1044-72-6). For the first starting period of operation, we will provide these for you.
  • Each tube should contain 3.2 pmol of a primer and a suitable amount of template. The total volume ( primer + template ) should be 3-5 ul.
  • The temperatures of your primers should be within the range of 55-65 degrees Celsius. Please let us know if your primers are outside of this range for some reason; we will need to perform sequencing PCR separately.
  • We strongly recommend checking both your template quality and concentration using agarose gel electrophoresis; spectrophotometer results are not good enough for estimating DNA quantity. (more info external link)
  • Use 100ng no-insert plasmid as a size/concentration marker for your plasmids whenever possible. For regular-sized plasmids (pBluescript, pUC18, etc.), run the gel with undigested plasmid.
  • Check the quality/integrity of your cosmids/PACs/BACs by EcoRI digestion and agarose electrophoresis.
  • Tubes (first and last in the strip) must be clearly labeled with numbers and your initials.

Naming Samples

Sample names are created by SeqCore. The names are uniform and describe the sequencing run only. The names will contain your initials, date, one letter, and sample number (e.g., BT_030807a01). If you would like to name your samples yourself, please e-mail us the names.

  • Names may include any combination of primer name, vector, date, number of sequence, etc. 
  • Characters: !'%&*$|/\{}[]()<>: and blank spaces are not to be used
  • 31-character limit 

Timing

Samples submitted by 6:00 pm Sundays will run Mondays, with results reported by Tuesday morning. Samples submitted by 6:00 pm Wednesdays will run Thursdays, with results reported by Friday morning.

Results

Results are typically e-mailed or transferred via FTP site. Please contact us for more information on how to use chromatogram files if needed.
