Electroporation of ES Cells, Clone Selection, and DNA Isolation

  1. Upon receipt of the linear DNA from the customer, the electroporation will be scheduled. Samples will be prioritized by the date received. 
  2. Customers will be notified of the electroporation date and the expected date for their receipt of DNA samples (see Timeline for approximate schedule). 
  3. DNA will be electroporated into ES cells (CJ7, R1, etc.) 
  4. After electroporation and selection, several hundred clones will be isolated, cultured in multi-well plates, and replicated. 
  5. One replicate plate will be placed in frozen storage and DNA will be isolated from each clone in the other replicate.
  6. The DNA samples will be shipped on dry ice to the customer for genotype analysis. Generally, the DNA will be delivered to the customer three weeks from the electroporation date.
  7. Customers are responsible for paying the FedEx shipping charges for the DNA samples.
  8. Since storage of frozen cells in multi-well dishes is not optimal, it is strongly suggested that customers return the genotype results to our laboratory within four weeks of receipt of the DNA samples.
